Regulation of InsP3R expression during cardiac hypertrophy
Postdoctoral Research Scientist

Starting salary in range £28,409 to £29,356 p.a.

The Babraham Institute is a charity which is world renowned for its research in life sciences to enhance lives and improve wellbeing.

We are seeking a talented Postdoctoral Research Scientist to study the regulation of expression of the inositol 1,4, 5 triphosphate receptor (InsP3R) intracellular calcium release channel by microRNA-133 during cardiac hypertrophy. This project aims to develop our previous findings that show that calcium signals arising from InsP3Rs contribute to the induction of cardiac hypertrophy (Higazi et al., 2009. Mol Cell) as well as to the pathology associated with it (Harzheim et al., 2009. PNAS), which place InsP3Rs as key regulators of myocyte function during health and disease.

The project will use cell and animal models of hypertrophy to dissect out the contribution of this miRNA pathway to regulation of InsP3R expression. Adenoviral approaches will be used to manipulate key regulators of hypertrophy-associated pathways in vitro and combinations of cell imaging, ChIP and qPCR to quantitate hypertrophic responses. Confocal imaging will be extensively used to assay calcium signalling mechanisms in primary cardiac myocytes and visualise translocation of GFP-tagged proteins.

Applicants must have a PhD in Cell Biology or Physiology. They must also have a working knowledge of signalling pathways, microRNA function and experience in calcium imaging as well as in cardiac physiology. They must also have experience in molecular biology and biochemical techniques as well as in good laboratory practice.
